چکیده

The authors have applied the thiochrome method for determining thiamine to histochemical investigations and could observe the localization of thiamine in cells and tissues (1-6). Independently of our study, v. Muralt (7) had already reported a investigation on histological pictures of thiamine in peri pheral nerves by the thiochrome method, which was however unavailable to the authors at that time. According to the retest of v. Muralt's method by the authors, preparations of peripheral nerves which can be observed comparatively well, were attained, but failed in case of organs containing much reducing substances such as the liver and kidneys. This paper deals with the procedure used at present by the authors and some histological pictures of main organs obtained by this method.

A histochemical study of the Golgi apparatus or 'acroblast' of the spermatid of the snail, Helix aspersa

The acroblast of Helix spermatids is a duplex structure: it consists of an 'externum' (which appears dark by positive phase-contrast microscopy and becomes impregnated with osmium) enclosing a region called the 'internum'. Histochemical studies show the externum to be composed principally of phospholipid and to contain glycolipid, alkaline phosphatase, and an enzyme which splits thiamine pyroph...

Three-dimensional Observations on Thick Biological Specimens by High Voltage Electron Microscopy

Thick biological specimens prepared as whole mount cultured cells or thick sections from embedded tissues were stained with histochemical reactions, such as thiamine pyrophosphatase, glucose-6-phosphatase, cytochrome oxidase, acid phosphatase, DAB reactions and radioautography, to observe 3-D ultrastructures of cell organelles producing stereo-pairs by high voltage electron microscopy at accere...

A Study of Acute and Chronic Anti-Nociceptive and Anti-Inflammatory Effects of Thiamine in Mice

Background: Thiamine (VitB1) is a vitamin with various important physiological functions and postulated therapeutic effects.  Its use as an analgesic in neuropathic pain has been undergoing in clinical settings. However, there has been little experimental investigation on this effect. In this study, anti-nociceptive and anti-inflammatory effects of thiamine were investigated in mice. Methods: T...

Further ultracytochemical analysis of rat adenohypophyseal cells: detection of two distinct enzyme activities within a single cell.

In an effort to ascertain whether a lysosomal enzyme, aryl sulphatase (ArSase), might share the same cell and, or, the same intracellular structure(s) with a histochemical Golgi apparatus marker, thiamine pyrophosphatase (TPPase), in rat pituitary mammotrophs and somatotrophs a technique for a selective demonstration of both enzyme activities within a single specimen has been developed. The tec...

Spectrophotometric Determination of Acidity Constants of Thiamine in Water, Water-Triton X-100 Micellar Media Solutions

In this work the acidity constants of Thiamine in water and water-Triton X-100 micelle media solutions, at 25 °C, have been determined spectrophotometrically. To evaluate the pH- absorbance data, a resolution based on the combination of soft-and hard-modeling is applied. The acidity constants of all related equilibria are estimated using the whole spectral fitting of the collected data to a...
